The hsa-mir-21_DDSD_s (BBa_K5086106) is formed by the combination of hsa-mir-21_DDSD (BBa_K5086012), hsa-mir-21_target (BBa_K5086002) and wt (BBa_K5086017) chains. Among them, the sequence at the 5' end of hsa-mir-21_DDSD is successively complementarily pair-linked to the hsa-mir-21_target and wt chains, and the sequence at its 3' end is complementary to the Universalness_h (BBa_K5086009) chain, so that hsa-mir- 21_DDSD_s can bind specifically to the Universalness_h strand thus accomplishing the recognition of the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction starting from the 3' end of the target hsa-mir-21_Rep. During DNA polymerization by Bst DNA polymerase, the wt strand is replaced and released by the newly synthesized daughter strand, which also releases a DNA single strand with the same sequence as the target strand. At the same time, a single strand of DNA with the same sequence as the target strand (U to T) will be released, which will serve as the target to undergo the first step of the polymerase-mediated strand displacement reaction again, realizing the DDSD self-enhancement process, and the wt strand will be involved in the subsequent reaction to produce the fluorescence report.
